Magugu studied fashion design, fashion photography and fashion media at LISOF in Johannesburg, with Mail & Guardian reporting that he graduated in 2014. He launched the eponymous THEBE MAGUGU label in Johannesburg in 2016 and is publicly described as its founder and director.
Magugu's ready-to-wear collections are presented as vehicles for storytelling and education, frequently engaging with South African history, culture, politics, family and ancestry. His label also publishes Faculty Press, an annual zine documenting contemporary South Africa, Africa and their diaspora through emerging cultural voices.
In 2019, Magugu won both the International Fashion Showcase and the LVMH Prize. The Victoria and Albert Museum identifies him as the first African designer to receive the LVMH Prize for emerging talent, making the award a significant point in his international recognition.
The label launched its first e-commerce site in 2020. Magugu later collaborated with Adidas on the Autumn/Winter 2022 sportswear collection Finding Beauty, and in May 2024 opened Magugu House in Johannesburg, described as his first physical retail space and showroom.
